Single-Cell Optical Nanomotion of Candida albicans in Microwells for Rapid Antifungal Susceptibility Testing

چکیده
Candida albicans is an emerging multidrug-resistant opportunistic pathogen representing important source of invasive disease in humans and generating high healthcare costs worldwide. The development a rapid simple antifungal susceptibility test (AFST) could limit the spread this disease, increase efficiency treatment, lower risk developing resistant strains. We developed microfluidic chip containing array microwells that were designed to trap cells perform tests using optical nanomotion detection (ONMD). Yeast cell entrapment microwell allows for very exchange growth medium with antifungal, which enables performing single-cell ONMD measurements on same before after treatment. exposure low concentration caspofungin or flucanozole induced significant decrease signal, demonstrating sensitivity technique. used quantify real-time response individual C. treatment as fast 10 min. This label-free technique be further into simple-to-use device performance AFST part routine hospital procedure also eventually world countries.

Standardized method for in vitro antifungal susceptibility testing of Candida albicans biofilms.
Candida albicans is implicated in many biomaterial-related infections. Typically, these infections are associated with biofilm formation. Cells in biofilms display phenotypic traits that are dramatically different from those of their free-floating planktonic counterparts and are notoriously resistant to antimicrobial agents. Consequently, biofilm-related infections are inherently difficult to t...
متن کاملMolecular identification and in-vitro antifungal susceptibility testing of Candida species isolated from patients with onychomycosis
Background and Purpose: Candida species are the most opportunistic fungi affecting the nails and resulting in onychomycosis. In this study, we identified and evaluated in-vitro susceptibility of the recovered isolates against fluconazole (FLC), voriconazole (VRC), and clotrimazole (CLT) using the Clinical and Laboratory Standards Institute (CLSI) M27-A3 document. Materials and Method...
متن کاملCollaborative study of the NCCLS and flow cytometry methods for antifungal susceptibility testing of Candida albicans.
One hundred clinical isolates of Candida albicans were tested for amphotericin B and fluconazole susceptibilities by the National Committee for Clinical Laboratory Standards (NCCLS) broth microdilution test at center 1 (C1). The same isolates were tested blinded at center 2 (C2) by NCCLS and flow cytometry (FC) methods. The agreement between NCCLS and FC methods ranged from 96 to 99%.
متن کاملPrevalence and Antifungal Susceptibility of Candida albicans in Patna, India
With changes in the medical and surgical management of patients over the past three decades, fungi have emerged as a major cause of human disease. Of the disseminated mycoses, candidiasis remains the most prevalent, with Candida albicans causing more invasive infections than any other fungus (Elizabeth, 2009). Candidiasis or thrush is a fungal infection (mycosis) of any of the Candida species, ...
متن کاملIncidence and Antifungal Susceptibility of Candida Albicans Infections
INTRODUCTION. Candida albicans is the most common inhabitant of the skin, mouth, vagina and gastro intestinal tract of human beings. One of the major reasons for the increase in Candida infection is the development of its resistant strains due to drugs used in the treatment of candidiasis. MATERIALS AND METHODS We studied 4027 samples collected from patients in various wards of the Emergency Co...
